Detection of Cymbidium Mosaic Virus (CYMV) on Vanda Plants

چکیده
Orchid plants are the members of Orchidaceae consisting of more than 25,000 species, which are distributed almost all over the world but more abundantly in the tropics. There are 177 genera, 1,125 species of orchids that originated in Thailand. Orchid plant collected from different nurseries showing Chlorotic and mosaic symptoms were observed on Vanda plants and it was suspected to infect with virus. So the symptomatic plants were tested for Cymbidium Mosaic Virus (CYMV), Odontoglossum ring spot virus (ORSV), Poty virus and Tomato Spotted Wilt Virus (TSWV) with Direct Antigen CoatingEnzyme Linked Immunosorbent Assay (DAC-ELISA) and further confirmed by Transmission Electron Microscopy (TEM). With the two methods CYMV and ORSV were detected positively from the suspected imported samples and low positive results were observed for Potex, Poty virus and Tomato Spotted Wilt Virus (TSWV).

متن کاملA novel approach for simultaneous detection of Citrus yellow mosaic virus and Citrus greening bacterium by multiplex polymerase chain reaction
A method of multiplex polymerase chain reaction (PCR) was developed for the simultaneous detection of Citrus yellow mosaic virus (CYMV) and fastidious Citrus greening bacterium, Candidatus Liberibacter asiaticus (CLa) in sweet orange trees. Initially total DNA from individual CLa and CYMV infected citrus plants were mixed and both pathogens were detected simultaneously by multiplex PCR. Subsequ...
متن کاملProduction of virus-free orchid Cymbidium aloifolium (L.) Sw. by various tissue culture techniques
Orchids are affected by many viruses resulting in poor growth, yield and quality, and an overall decline in population. Cymbidium mosaic virus (CymMV) is one of the common orchid viruses found in Cymbidium species but it infects different orchid genera. In this study Cymbidium aloifolium was propagated in vitro using MS medium at different strength (1.0, ½, and ¼) with or without 0.5 mg/l BAP (...
متن کاملReciprocal function of movement proteins and complementation of long-distance movement of Cymbidium mosaic virus RNA by Odontoglossum ringspot virus coat protein.
Complementation of movement and coat proteins of the orchid-infecting potexvirus Cymbidium mosaic virus (CymMV) and tobamovirus Odontoglossum ringspot virus (ORSV) was investigated. Nicotiana benthamiana, which is susceptible to both CymMV and ORSV, was used as a model system. Four transgenic lines, each harbouring one of the movement protein (MP) or coat protein (CP) genes of CymMV or ORSV, we...
متن کاملDetection of two orchid viruses using quartz crystal microbalance (QCM) immunosensors.
Quartz crystal microbalance (QCM) immunosensors are based on the principle that adsorption of substances on the surface of a quartz crystal changes its resonance oscillation frequency. A QCM immunosensor was developed for the detection of both cymbidium mosaic potexvirus (CymMV) and odontoglossum ringspot tobamovirus (ORSV) by pre-coating the QCMs with virus-specific antibodies. Upon binding of...
